Silymarin and its components can protect cardiac microsomes and mitochondria from adriamycin-induced lipid peroxidation in rats, suggesting that silymarin can prevent adriamycin-mediated cardiotoxicity. Silymarin has been found in animal models to protect the heart from ischemia-reperfusion injury, possibly through preconditioning.Evidence seems to suggest that silymarin components reduce the activity of ERK /MEK and IP3K/Akt survival promoting pathways, activation of which is central to pretreatment of Ah, bradykinin, and Ouabain.It is also important that the silymarin component stimulates estrogen receptors and inhibits matrix metalloproteinases, phosphodiesterase and mitochondrial ROS production to facilitate preconditioning.In addition, the anti-inflammatory properties of certain ingredients may play a role in protecting tissue.Although the primary silybin is generally suspected to have these effects, other minor components of the extract have also been shown to have important cardioprotective activity.
